Nanozyme technology at Moscow State University. Achievements and development perspectives

The work describes novel functional bionanosystems for treatment and diagnostics on the basis of proteins, enzymes, polymeric coatings, and magnetic nanoparticles developed at Lomonosov Moscow State University Laboratory for Chemical Design of Bionanomaterials in collaboration with scientists from UNC Eshelman School of Pharmacy (USA). The properties of enzymes (superoxide dismutase, catalase, organophosphate hydrolase, and lysines of bacteriophages) and other drug molecules immobilized in polymeric complexes, as well as the methods for targeted drug delivery using cell-mediated systems and magnetic nanoparticles in in vitro and in vivo operating conditions, are discussed. Physical and chemical characteristics, including data on the functional properties of the nanoformulations, are obtained. The nanoformulations developed demonstrated high potential therapeutic efficacy for the treatment of central nervous system and brain diseases, inflammations (including inflammatory diseases of the eye), cancer and infectious diseases, neurotoxic injury, and others. The possibilities of remote control biochemical reactions using a nonheating low-frequency alternating magnetic field (AMF) for the controlled release of drugs are analyzed in the review. The experimental results of the AMF effects on bionanosystems containing magnetic nanoparticles, such as changing the catalytic activities of enzymes bound to magnetic nanoparticles and ‘disordering’ of the lipid bilayer in membranes, are considered.

Preparation of the recombinant НU-proteins from S. melliferum and M. gallisepticum and of their complexes with DS-DNA for structural NMR experiments

We created a high-performance system for expression of 13C, 15N-double-labeled recombinant НU-proteins from mycoplasms S. melliferum (HUSpm) and M. gallisepticum (HUMgal), and a purification protocol that yields protein samples suitable for structure studies by high-resolution NMR. Using these proteins we optimized the conditions of NMR experiments and obtained two-dimensional heteronuclear 1H/15N-HSQC and 1H/13С-HSQC-CT NMR spectra of free НU-proteins (HUSpm and HUMGal), and 1H/15N-HSQC spectrum of a complex between HUSpm and a double-stranded DNA (ds-DNA). The techniques we created are applicable in the studies of НU-proteins from other organisms. They allow analyzing structure and dynamics of free proteins and of their conformational changes induced by binding of various ds-DNAs or of DNA binding inhibitors.

Tetrameric RGD induces clustering of integrin αvβ3 on the melanoma cell surface and decreases cell viability

摘要

The recombinant hybrid protein SR15 composed of streptavidin fused with RGD peptide is obtained. The ability of this protein to recognize human melanoma cells (MeWo line) is demonstrated. Types of expressed RGD-binding integrins in the cells of this line are identified. It is found that the recombinant hybrid protein SR15 binds to integrin αvβ3 on the surface of human melanoma cells. The binding to the protein SR15 results in clustering of αvβ3 receptors on the surface of MeWo cells, internalization of the recombinant protein, and an almost twofold decrease in cell viability.

Screening of bacterial genes responsible for resistance to beta-lactam antibiotics using microarrays with enzymatic detection

The method of hybridization analysis on microarrays with enzymatic detection based on horseradish peroxidase is applied to screen infectious agents of nosocomial and community-acquired infections for beta-lactamase genes causing resistance to beta-lactam antibiotics. The advantages of using this method for the rapid identification of genes are demonstrated. Similarities and differences in the distribution of beta-lactamase genes identified in the infectious agents of nosocomial and community-acquired infections are revealed. The most common type of extended-spectrum beta-lactamases (ESBLs) is CTX-M. The high prevalence of extended-spectrum beta-lactamases, particularly of the TEM-1 beta-lactamase, is demonstrated. Individually or in combination with genes of TEM-1 and SVH-1 beta-lactamases, the genes of subgroup CTX-M-1 beta-lactamases were the most frequently identified in community-acquired infectious agents. There were no cases of the simultaneous detection of multiple ESBLs in community-acquired infectious agents. Much more varied combinations of beta-lactamases were identified in nosocomial infectious agents: a combination of extended-spectrum beta-lactamases and broad-spectrum beta-lactamases was identified in 62% of strains and the simultaneous presence of two different types of ESBLs was identified in 18% of strains.

Influence of Met/Leu amino acid changes on catalytic properties and oxidative and thermal stability of yeast D-amino acid oxidase

The oxidative stability of enzymes is mostly dependent on the stability of the Cys and Met residues. Three single point mutants with Met/Leu substitutions in D-amino acid oxidase (DAAO, EC 1.4.3.3) from the yeast Trigonopsis variabilis (TvDAAO) are prepared and characterized. The positions for the amino acid residue substitutions are selected based on multiple alignment of different DAAO amino acid sequences and analysis of the three-dimensional structure of TvDAAO. It is shown that the substrate specificity profile ischanged for all of the mutants. The KM values for the small and bulky D-amino acidsare increased and decreased, respectively. One of the Met/Leu substitutions results in a two- to threefold increase in thermal stability as compared to the wild-type enzyme. A method for the determination of TvDAAO stability in the presence of hydrogen peroxide is developed and the oxidative stability of wild-type and mutant TvDAAOs is studied. It is shown that none of thethree mutations changes the oxidative stability of the enzymes.

A new sampling format for the diagnostics of bovine infectious diseases in dried blood spots by ELISA

The detection of antibodies to bovine infectious diseases (enzootic bovine leucosis, viral diarrhea, and infectious rhinotracheitis) is performed by ELISA in dry whole blood samples obtained on porous membranes using a new sampling format. Several commercial test systems are used to diagnose the infections. The obtained data are compared to the results of the analysis of liquid samples of blood serum. The results of the analysis of dried and liquid samples are in full concordance. The new sampling format makes it possible to avoid using a cold chain for the transportation of samples to diagnostic laboratories and to simplify the storage of these samples.

The relationship between NO-synthase inhibitory activity of N,S-containing heterocycles and their radioprotective and antileukemic properties

The effect of NO-synthase (NOS) activator and inhibitors on leukemic cell lines HL-60, K-562, and MOLT-4 and bone marrow cells of untreated patients diagnosed with B-cell acute lymphoblastic leukemia compared with lymphocytes from healthy donors is examined. The obtained data on the relationships between the radioprotective, NOS inhibitory, and cytotoxic properties of a number of thiazine, thiazoline, and thiourea derivatives indicates their potential for use as agents for complex radio- and chemotherapy.

Electron spectra of WO molecule. New electron state 3Π

摘要

The electron spectrum of the tungsten monooxide molecule is observed in the 550–800 nm region using intracavity laser absorption spectroscopy. The WO molecules are produced in a pulsed electric discharge through the mixture of tungsten hexacarbonyl vapors. The spectrum is recorded using a diffraction spectrometer (resolving power of 240000). The bands in the 16400–15500 cm–1 region are assigned to the 3Π0–X3Σ+ component of the 3Π0–X3Σ+3+ electron transition. The rotational analysis of the 0-0 and 1-0 bands is carried out and the rotational constants for the ground X″3Σ and the exited 3Π0 states are computed: В′ = 0.385738 cm–1, B″ = 0.415538 cm–1.

Cobalt(II) porphyrazine as an active component of iodide-selective electrodes

Cobalt(II) porphyrazine is synthesized and studied as an active component of a polyvinyl chloride plasticized membrane ion-selective electrodes (ISEs). It is established that regardless of their structure, ISEs are sensitive to iodide. The introduction to the ISE of an ionic additive, ionic liquid 1,3-dihexadecylimidazolium chloride, significantly improves the electrochemical characteristics: the slope of the electrode function reaches −(57 ± 1) mV/dec, cmin = 8.3 × 10–6 M. Solid-state screen-printed electrodes the surfaces of which are modified by a 1: 4 mixture of cobalt(II) porphyrazine and ionic liquid 1,3-dihexadecylimidazolium chloride demonstrate satisfactory electrochemical characteristics: the slope of the electrode function is −(56 ± 4) mV/dec and cmin = 2.5 × 10–5 M. The potentiometric selectivity of the ISEs for iodide is studied. It is found that the effect of lipophilic interfering ions is significantly lower for solid state ISEs than for plasticized membrane electrodes.

Using the size-exclusion chromatography method, the molecular-mass distribution of dissolved organic matter of drinking water and water sources of Ufa is studied. The influence of seasonal changes and various water treatment stages on the molecular-mass distribution of dissolved organic matter is shown.
